It can - but it has to run under emulation (DOSBox, Virtual PC or VMware).

If you purchase the game from GOG - it will include DOSBox (as well as setting it up automatically to run under it).

Dear Fellow detectives,

I am having a fit over installing Tex Murphy Under a Killing Moon in Vista. I already downloaded DOSbox, but can't get it to install 'Moon'. I tried to mount the 4 Tex files with Daemon Tools but it won't work. Does anybody have a simple solution for an exasparated wannabe detective who's terrible with computers?

Have you set up DosBox correctly? You need to mount folders or drives in DosBox for it to recognize them. There's a guide for it here.
